Indications:

🌿 Menopausal Vasomotor Symptoms

  • Indicated for the treatment of moderate to severe vasomotor symptoms (e.g., hot flashes, night sweats) associated with menopause.

🌿 Prevention of Postmenopausal Osteoporosis

  • Used for the prevention of osteoporosis in postmenopausal women at high risk who are intolerant of or contraindicated for non-estrogen therapies.

🌿 Estrogen Deficiency (Hypoestrogenism)

  • Indicated for estrogen replacement in women with ovarian failure, hypogonadism, or after oophorectomy when systemic estrogen therapy is required.
Dosage & Administration:

✅ Treatment of Estrogen Deficiency Symptoms (Menopausal Vasomotor Symptoms, Vaginal Atrophy):

  • Starting dose: 1.25 g of gel once daily (delivering 0.75 mg estradiol per day).
  • Dose adjustment: Based on clinical response; may be increased up to 2.5 g daily (delivering 1.5 mg estradiol per day).
  • Apply once daily to clean, dry, unbroken skin on the upper arms, shoulders, or inner thighs.

Instructions for Use:

  • Spread the gel thinly over the recommended area; do not massage into the skin.
  • Allow to dry for 5 minutes before dressing.
  • Avoid washing the application site for at least 1 hour after application.
  • Rotate application sites to reduce skin irritation.
Mode of Action:

Estradiol, the active ingredient in Oestrogel, is a form of endogenous estrogen that binds to estrogen receptors in target tissues, such as the reproductive system, bone, breast, and brain. It regulates gene expression and protein synthesis to restore estrogen levels, reducing vasomotor symptoms (hot flashes), preventing bone loss, and improving vaginal atrophy associated with estrogen deficiency in postmenopausal women.

Drug Interactions:

✅ Estradiol may interact with:

  • Enzyme inducers (e.g., rifampicin, carbamazepine, phenytoin, St. John’s Wort) → may reduce estradiol effectiveness by increasing its metabolism.
  • Enzyme inhibitors (e.g., erythromycin, ketoconazole) → may increase estradiol levels and risk of side effects.
  • Thyroid hormones → may alter thyroid-binding globulin levels; monitoring may be needed.
  • May increase effects of corticosteroids, cyclosporine, and theophylline by reducing their clearance.

✅ Caution when used with anticoagulants or antidiabetic drugs due to possible alterations in their effects

Contraindications:

Oestrogel is contraindicated in patients with:
✅ Known, suspected, or history of breast cancer or estrogen-dependent tumors
Undiagnosed abnormal genital bleeding
Active or history of thromboembolic disorders (e.g., deep vein thrombosis, pulmonary embolism)
Active or recent arterial thromboembolic disease (e.g., stroke, myocardial infarction)
✅ Known liver dysfunction or liver disease
Porphyria
✅ Known hypersensitivity to estradiol or any component of the gel
